1.2.8 Bioinformatics Analysis


SDSC BIOLOGY WORK BENCH is a tool kit which provides easy access for various bioinformatics tools to analyze the data. CLUSTAL W tool a product of SDSC was used to perform multiple sequence analysis of the TPI protein among different organisms. Phylogenetic study was made using the DENDROGRAM (Fig. 1.2). TEXSHADE and BOXSHADE were used to identify the common conserved regions among the sequences (Figs. 1.3, 1.4).

The dendrogram indicates that query and Lactobacillus delbrueckii fall under common branch which shows that they share maximum similarity where as Homo sapiens falls under a completely different branch which indicates there evolutionary divergence

1.3 Results and Discussion


Different Lactobacillus strains will produce bacteriocins but the Lactobacillus delbrueckii strain is mostly used for the production of more amount of Bacteriocin as the TPI gene is only present in the Lactobacillus delbrueckii strain so the Lactobacillus delbrueckii is isolated from raw cow milk and the Lactobacillus delbrueckii strain were identified by several biochemical tests and the confirmatory test was mannitol test which shows no acid and no gas production which confirms the presence of the Lactobacillus delbrueckii and genomic DNA was isolated by proteinase K method and its TPI gene was amplified by using PCR. The Bacteriocin produced by the Lactobacillus delbrueckii is used as preserving agent in dairy and food industries and it used to prepare probitics to treat several diseases caused by bacteria. The Bacteriocin is used in the preparation of sporlac powder which is used to treat diarrhea. The Bacteriocin produced by the Lactobacillus strains is called lacticin which is mainly used in the food preserving and diary industries.
